Gd(III)‐19F Distance Measurements of Proteins in Cells by Electron‐Nuclear Double Resonance

چکیده
Studies of protein structure and dynamics are usually carried out in dilute buffer solutions, conditions that differ significantly from the crowded environment cell. The double electron-electron resonance (DEER) technique can track proteins’ conformations cell by providing distance distributions between two attached spin labels. This technique, however, cannot access distances below 1.8 nm. Here, we show GdIII-19F Mims electron-nuclear (ENDOR) measurements cover part this short range. Low temperature solution in-cell ENDOR measurements, complemented with room PRE (paramagnetic relaxation enhancement) NMR were performed on fluorinated GB1 ubiquitin (Ub), spin-labeled rigid GdIII tags. proteins delivered into human cells via electroporation. derived essentially identical lie 1–1.5 nm range revealing both, Ub, retained their overall 19F regions

DEER distance measurements on proteins.
Distance distributions between paramagnetic centers in the range of 1.8 to 6 nm in membrane proteins and up to 10 nm in deuterated soluble proteins can be measured by the DEER technique. The number of paramagnetic centers and their relative orientation can be characterized. DEER does not require crystallization and is not limited with respect to the size of the protein or protein complex. Diama...
متن کاملImproved Sensitivity for Long-Distance Measurements in Biomolecules: Five-Pulse Double Electron–Electron Resonance
We describe significantly improved long-distance measurements in biomolecules by use of the new multipulse double electron-electron spin resonance (DEER) illustrated with the example of a five-pulse DEER sequence. In this sequence, an extra pulse at the pump frequency is used compared with standard four-pulse DEER. The position of the extra pulse is fixed relative to the three pulses of the det...
متن کاملPulse Dipolar Electron Spin Resonance: Distance Measurements
In recent years electron spin resonance (ESR) has provided the means to obtain structural constraints in the field of structural biology on the nanoscale by measuring distances between paramagnetic species, which usually have been nitroxide spin-labels. These ESR methods enable the measurement of distances over the wide range from ca. 6–10 Å to nearly 90 Å. While cw methods may be used for the ...
متن کاملTheory of double quantum two-dimensional electron spin resonance with application to distance measurements
A formulation is presented for calculating double quantum two dimensional electron spin resonance ~DQ-2D ESR! spectra in the rigid limit that correspond to recent experimental DQ-2D ESR spectra obtained from a nitroxide biradical. The theory includes the dipolar interaction between the nitroxide moieties as well as the fully asymmetric g and hyperfine tensors and the angular geometry of the bir...
متن کاملAccurate long-range distance measurements in a doubly spin-labeled protein by a four-pulse, double electron-electron resonance method.
Distance determination in disordered systems by a four-pulse double electron-electron resonance method (DEER or PELDOR) is becoming increasingly popular because long distances (several nanometers) and their distributions can be measured. From the distance distributions eventual heterogeneities and dynamics can be deduced. To make full use of the method, typical distance distributions for struct...
